"I'm sick of it affecting my family and other families. Enough is enough, and let's take a stand," the actor, 41, tells PEOPLE.
After losing his father to bladder cancer when he was 5 and supporting his mother, who was diagnosed in 1998 and beat it, Daly was ready to do whatever it took to find a cure.
"Cancer is a very important part of my family as it is many people's. We've made a lot of progress, but there is still a lot of work to be done," explains The Voice host, who joined Stand Up to Cancer years ago when the telethon began.
The organization is part of a groundbreaking movement to accelerate innovative cancer research that will get new therapies to patients quickly – and hopefully save lives.
"Hollywood can get a bad rap, but this really is a great portrait of what this town can do," he says. "It's titans of various industries coming together to make a dent."
And recently becoming a father for the third time has only strengthened his resolve to find a cure.
"It really hits home when you have your own young family who are going to be a part of those staggering numbers, unfortunately," says Daly, who is father to Jackson, 5, Etta, 2, and newborn London. "This cancer fight is for my kids and their generation."
The biennial Stand Up to Cancer telethon airs Sept. 5 (8 p.m. ET) on ABC, CBS, FOX, NBC and several other networks
