Farrell, 38, and Vaughn, 44, had been rumored to be cast in the leading roles for several weeks.
In its announcement, the network also revealed that the new season of the gritty drama will be eight one-hour episodes and offered this description: "Three police officers and a career criminal must navigate a web of conspiracy in the aftermath of a murder."
Farrell will play Ray Velcoro, a compromised detective whose allegiances are torn between his masters in a corrupt police department and the mobster who owns him. And Vaughn will star as Frank Semyon, a career criminal in danger of losing his empire when his move into legitimate enterprise is upended by the murder of a business partner.
Justin Lin, who has directed films in the Fast and Furious franchise, will helm the first two episodes.
Matthew McConaughey and Woody Harrelson starred as the detectives in the first season of the HBO series, which garnered critical praise and a handful of Emmy nominations at this year's awards.
